The earliest known routing of route 44 is from early 2000, when the route traveled from 48th St & Baseline Rd, presumably turning right at Broadway Rd and going up Mill Ave to University Dr, before heading over to 44th Street (Arizona State Route 153 at the time), and heading up to Paradise Valley Mall. On March 19, 2000, the route was modified to instead travel from College Ave & 5th St in Downtown Tempe, down to University Dr and over to 44th St, up to Paradise Valley Mall, then continuing on to the Phoenix Mayo Hospital. On March 19, 2001, the northern end of the route was extended to Tatum Blvd & Deer Valley Rd. On January 14, 2002, the north end of the route was slightly adjusted to better serve Desert Ridge Marketplace, then on February 24, 2003, the north end of the route was slightly adjusted again, this time to better serve the Desert Ridge Marriott Resort. On July 27, 2009, the southern end of route 44 was truncated from Tempe Transit Center to University Dr & Hohokam Expressway (Arizona State Route 143). Finally, on January 28, 2013, the southern end was truncated again to the 44th St Sky Train Station.
